ABSTRACT

AIM:To investigate the effect of Shenci capsule combined with cisplatin ( DDP) in reversing DDP resistance by PI3K/AKT/mTOR signaling pathway in nude mice bearing A 549/DDP tumor.METHODS:The patient-de-rived lung adenocarcinoma A 549/DDP cell xenograft model was established .The tumor-bearing nude mice were randomly divided into control group , Shenci capsule group , DDP group and Shenci capsule combined with DDP group .The mice in control group was treated with normal saline , while the mice in other groups were treated with different drugs for 21 d.After treatment, the mice were killed and lung cancer tissues were collected .Flow cytometry was used to analyze the cell cycle and apoptosis.FQ-PCR was used to determined the mRNA levels of PTEN , P-glycoprotein, PI3K, AKT and mTOR in A549/DDP lung tumor .RESULTS:Compared with control group , the cell proliferation in all the drug treatment groups was inhibited .Compared with other drug treatment groups , Shenci capsule combined with DDP blocked the cell cycle of A 549/DDP cells at G2/M phase, promoted the apoptosis rate , increased the mRNA expression of PTEN and inhibited the mRNA expression of P-glycoprotein, PI3K, AKT and mTOR.CONCLUSION:Shenci capsule increases the sensitivity of A 549/DDP resistant cells in nude mice to DDP by blocking PI 3K/AKT/mTOR signaling pathway , increasing the expression of PTEN or inhibiting P-glycoprotein-mediated resistance pathway .

ABSTRACT

Objective To observe Shenci capsule's effect on immunologic function and hematological tox-icity of patients with advanced non-small cell lung cancer.Methods 60 cases of patients with advanced non-small cell lung cancer were chosen and divided into the therapy group and the control group randomly with 30 cases in each group respectively. While patients in the control group were administered simple GP chemotherapy regime, patients in the therapy group were administered Shenci capsule treatment orally in addition to the therapy in the control group.Comparisons were made of the variations in the subgroup of T lymphocyte(including the absolute val-ue of CD3+cell,the absolute value of CD4+cell,the absolute value of CD8+cell and CD4+/CD8+ratio)of hematology before and after chemotherapy between the two groups of patients. Results No significant discrepancy was found in the comparison of the absolute value of CD3+cell,the absolute value of CD4+cell,the absolute value of CD8+cell and CD4+/CD8+ratio before the chemotherapy between the two groups(P > 0.05)and the absolute value of CD3+cell,and the absolute value of CD4+cell of both groups decreased significantly after the chemotherapy(P<0.05), especially in the control group(P < 0.05). The hematological toxicity of the therapy group was lower than that in the control group(P < 0.05)after the chemotherapy. Conclusion Shenci capsule was capable of enhancing the body immunity in advanced non-small cell lung cancer and alleviating hematological toxicity.

ABSTRACT

Objective To build a method for determination of berberine, epiberberine, coptisine and palmatine in Shutongantai Capsules by HPLC.Methods Thermo C18 column (5μm, 4.0 mm× 250 mm) was used with acetonitrile-0.05 mol/L potassium dihydrogen phosphate solution (50∶50, 0.4 g sodium dodecyl sulfate was added in each 100 mL solution, to adjust the pH 4.0 with phosphoric acid) as mobile phase;the flow rate was 1.0 mL/min;detection wavelength was 345 nm;column temperature was 30℃.Results Berberine hydrochloride showed a good linear relationship in the range of 0.180 34-0.901 68μg (r=0.999 8).Conclusion The method is simple, accurate and reproducible, and can be used for the quality control of Shutongantai Capsules.

ABSTRACT

Objective To establish a quality standard of Shutongantai Capsules. Methods Thermo C18 column was used with methanol-0.1% phosphoric acid as mobile phase. The flow rate was 1.0 mL/min, detection wavelength was 254 nm, and column temperature was 30 ℃. Results Emodin showed a good linear relationship in the range of 0.016-0.080 μg (r=0.999 6). Chrysophanol showed a good linear relationship in the range of 0.032-0.160 μg (r=0.999 8). Conclusion The established method is simple, feasible and reproducible, and can be used for the quality control of Shutongantai Capsules.

ABSTRACT

Fuantai-03(FAT-03), isolated from the Dasyatis akajei, has a strong antiangiogenic activity. The recombinant Fuantai-03 (GST/rFAT-03) fusion protein can be obtained with the DNA recombination technology. In this study, expression conditions of GST/rFAT-03 were optimized by response surface experimental design method. The constructed engineering bacteria containing GST/rFAT-03 plasmid was induced by isopropy-beta-D-thiogalactosid (IPTG), the GST affinity column was used for isolation and purification, and then the effects of different culture time, IPTG concentration, induction temperature and induction time on the amount of soluble GST/rFAT-03 fusion protein were compared. The culture time for optimal expression was 6.13 h, IPTG concentration was 0.36 mmol/L, induction temperature was 19.71 degrees C, and induction time was 13.60 h. The amount of soluble GST/rFAT-03 fusion protein was 7.57 mg/L under above mentioned expression conditions. The results also showed that rFAT-03 significantly inhibited angiogenesis in chicken chorioallantoic membrane in a dose-dependent manner. Moreover, the soluble form of the target protein is useful for further work on purification and on studying its biological function.

ABSTRACT

@#Objective To observe the effects of electro-acupuncture and functional training on gross motor function of children with cerebral palsy. Methods 137 children with cerebral palsy were divided into observation group (n=81) and control group (n=56). The observation group received electro-acupuncture and functional training, and the control group received functional training only. Results The scores of Gross Motor Function Measure (GMFM) increased in both group after treatment (P<0.01), and it was significantly higher in the observation group than in the control group (P<0.01). Conclusion Electro-acupuncture and functional training is superior to functional training in gross motor function.

ABSTRACT

Objective To study the potential antidepressive-like effect of tetramethylpyrazine (TMP). Methods Forced-swimming and chronic mild stress (CMS) tests were performed to assess the antidepressant-like activity of TMP. Male Sprague-Dawley (SD) strain rats were divided into six matched groups (n= 13 or 14 in each group) based on their sucrose consumption:control, CMS, CMS + fluoxetine, and CMS + TMP groups. The rats except control were housed separately in different rooms, and the rat model of depression was established by exposing to an unpredictable sequence of stressors for 28 days; the rats in CMS + fluoxetine were exposed to CMS and received administration of FLU (2.0 mg· kg-1·d-1 ,ig) for 28 days; the rats in CMS + TMP groups were exposed to CMS and received administration of TMP (10,20,40 mg·kg-1·d-1 ,ig) , respectively, for 28 days. The rats in control group were given ordinary daily care and received ig administration of normal saline simultaneously. The body weight, food intake and fluid consumption were measured, and the behaviors were examined by open field during the duration of the stress procedure, and forced-swimming test was performed 1 day after last unpredictable stressor. Results Acute administration of TMP markedly decreased the duration of immobility during forced-swimming test((89.0 ±37.0)s vs (117.1 ±32. 1)s, P<0.05) . Chronic administration of TMP partially countered the effects of CMS on consumption of sucrose solution and locomotion and exploration behavior, and potently shortened the immobility time during forced-swimming test following CMS in rats. The results showed that long-term administration of TMP partially reversed the effects of CMS on the body weight gain,the consumption of sucrose solution,the squares crossing in open field test and the immobility time during forced-swimming test in rats ((91.9 ±31.5) vs (124.4±27.0)s,P<0.05).Conclusion TMP shows obvious antidepressant-like activity.

ABSTRACT

Objective To investigate the effects of SDY-08 isolated from Dasyatis akajei on tis-sue repair. Methods MTT assay was performed to measure the effect of SDY-08 on the growth of ECV-304 cells. The effect of SDY-08 on angiogenesis was detected in the chick embryochorioallantoic membrane (CAM). Mouse wound model was applied to investigate the effect of SDY-08 on tissue repair. Immunohistochemical staining assay and Western blotting were adopted to examine the expression changes of vascular endothelial growth factor (VEGF), Bcl-2 and Bax in wound tissues in response to SDY-08. Results The proliferation rates of SDY-08 at final concentration of 80 μg/ml promoting ECV-304 cells were 28.1%, 115.6% and 81.4% respectively at 12, 24 and 36 hours. The induction rate of angiogene-sis of CAM by SDY-08 at concentration of 1.6 mg/ml was 72.1%. SDY-08 at 0.5 mg/ml markedly in-duced acceleration of wound healing in mouse model two days in advance. SDY-08 up-regulated either ex-pressions of VEGF in trauma group or that of Bcl-2 and VEGF of ECV-304 cells, but down-regulated ex-pression of Box. Conclusions SDY-08 can significantly promote angiogenesis and tissue repair, which is closely correlated with its effect of up-regulating expressions of VEGF and Bcl-2 as well with that of down-regulating expression of Box.

ABSTRACT

AIM To investigate whether tubeimosides induce apoptosis in CNE-2Z. METHODS Growth inhibition by tubeimosides was measured using MTT assay. The effect of tubeimosides on apoptosis induction of CNE-2Z cell line was studied by the fluorescent microscopy, electronic microscopy, DNA agarose gel electrophoresis, flow cytometry analysis. Western blotting was performed for detecting the apoptosis-related gene. RESULTS The growth of CNE-2Z cells was inhibited obviously by tubeimosides. CNE-2Z cells showed typical apoptotic features observed by fluorescent microscopy, electronic microscopy. DNA fragmentation into multiples of low molecular weight DNA(180~200 bp) was detected by agarose gel electrophoresis of DNA; Sub-G 1 peak was found by flow cytometry analysis, and the apoptosis index was 58 0%. The expression of bcl-2, inhibitor of apoptosis, decreased apparently; while that of bax, inducer of apoptosis, increased notably at 1, 3, 5 h after the addition of tubeimosides. The cleavage of caspase-3 was noticed at 1, 3, 5 h after the treatment of tubeimosides. CONCLUSION Tubeimosides could induce the apoptosis of CNE-2Z cells. The induction of apoptotic by tubeimosides was closely associated with bcl-2, bax and caspase-3.

ABSTRACT

Objective To study the effect of active ingredients(saikosaponin) of bupleurum chinensis D C.on GABA of hippocampus of chronic kindling induced by pentylenetetrazol(PTZ) in rats.Methods Forty-eight healthy Sprague-Dawley rats were randomly divided into six groups with eight in each group: normal control(Group A), normal saline(NS) group(Group B),sodium valproate(VPA) group(Group C),group of large-dose saikosaponin(Group D),group of medium-dose saikosaponin(Group E),and group of small-dose saikosaponin(Group F).Except Group A to be controlled,the other groups were given different treatments when chronically kindled by PTZ.The process of the test lasted for 4 weeks.After 4 weeks' administration the brain tissues were sampled,sliced and stained by enzyme immunohistochemistry(EIH).The results were analyzed according to the positive cell population and gray value.Results The positive cell population of CA1,CA2 and DG in group B was significantly different from that in the other groups(P
